How GeniePlex® Works
GeniePlex® is based on a bead-based sandwich immunoassay. Different bead populations are distinguished according to their size and fluorescence intensity, with each population conjugated to a specific capture antibody.
After the target analytes are captured, a biotinylated detection antibody binds to a second epitope, followed by detection with Streptavidin-R-Phycoerythrin (PE). The flow cytometer identifies each bead population and measures the corresponding PE signal. Analyte concentrations are determined by comparison with a standard curve, allowing multiple biomarkers to be quantified simultaneously from the same sample .
GeniePlex® Workflow
| 1 | Capture – 60 min: incubate samples with capture antibody-conjugated beads at room temperature with shaking, followed by washing |
| 2 | Detection – 30 min: add the biotinylated detection antibody, incubate at room temperature with shaking and wash |
| 3 | Signal development – 20 min: add Streptavidin-PE and incubate at room temperature with shaking |
| 4 | Flow cytometry analysis: wash, resuspend the beads in Reading Buffer and acquire the samples on a compatible flow cytometer |
The complete GeniePlex® workflow can be completed in approximately 4 hours, enabling multiple biomarkers to be evaluated in a single experiment instead of running numerous individual ELISA tests.
Required Equipment
In addition to a compatible flow cytometer, the GeniePlex® workflow requires a filter plate washer (vacuum manifold) for the wash steps and a plate shaker. Assay Genie recommends the GeniePlex® Filter Plate Washer NX.
